Enterokinase is a protease of the intestinal brush border that specifically cleaves the acidic propeptide from trypsinogen to yield the active trypsin. It cleaves the sequence DDDK, the substrate binding site RRRK differs from the bovine EK site by one amino acid and this is thought to be accountable for the much higher specific activity. It is composed of two domains, a heavy chain and a light chain. The 235 amino acid light chain is the catalytic domain.
