DTSSP is a cleavable protein crosslinking reagent that contains a disulfide moeity in its eight atom spacer arm. DTSSP is a membrane impermeable, homo-bifunctional crosslinker that is reactive towards amine (amino) functional groups. In the pH range of 7-9, this reaction is particularly selective for primary amine groups, resulting in the formation of stable amide linkages. Phosphate buffers & phosphate buffered saline are typically used for crosslinking reactions involving DTSSP, although other non-amine containing buffers may also be used.
